酵素学においてイソプレン合成酵素（イソプレンごうせいこうそ、イソプレンシンターゼ、isoprene synthase、EC 4.2.3.27）は、以下の化学反応を触媒する酵素である。
ジメチルアリル二リン酸 ⇌{displaystyle rightleftharpoons }
酵素の基質はジメチルアリル二リン酸であり、2つの生成物はイソプレンおよび二リン酸である。
この酵素はリアーゼファミリー、具体的にはリン酸エステルに作用する炭素-酸素リアーゼに属する。本酵素群の系統名はdimethylallyl-diphosphate diphosphate-lyase (isoprene-forming)である。一般的にはISPC、ISPSの略称が使われる。
